S'all about generating transgenics. A lot of it is trying to replicate the endogenous expression pattern of a particular locus, but using a reporter for visualisation. It's a question of where it is expressed, when, how much, in response to such and such etc.

Quite often there are regulatory elements in the downstream area, even quite far away.
